Thioredoxin-1 maintains the cellular redox status and decreases the infarct size in ischemia/reperfusion injury. However, whether the increase of thioredoxin-1 expression or its lack of activity modifies the protection conferred by ischemic postconditioning has not been yet elucidated. The aim was to evaluate if the thioredoxin-1 overexpression enhances the posctconditioning protective effect, and whether the lack of the activity abolishes the reduction of the infarct size. Wild type mice hearts, transgenic mice hearts overexpressing thioredoxin-1, and a dominant negative mutant (C32S/C35S) of thioredoxin-1 were used. The hearts were subjected to 30 min of ischemia and 120 min of reperfusion (Langendorff) (I/R group) or to postconditioning protocol (PostC group). The infarct size in the Wt-PostC group decreased in comparison to the Wt-I/R group (54.6 ± 2.4 vs. 39.2 ± 2.1%, p < 0.05), but this protection was abolished in DN-Trx1-PostC group (49.7 ± 1.1%). The ischemia/reperfusion and postconditioning in mice overexpressing thioredoxin-1 reduced infarct size at the same magnitude (35.9 ± 2.1 and 38.4 ± 1.3%, p < 0.05 vs. Wt-I/R). In Wt-PostC, Trx1-I/R and Trx1- PostC, Akt and GSK3β phosphorylation increased compared to Wt-I/R, without changes in DN-Trx1 groups. In conclusion, given that the cardioprotection conferred by thioredoxin-1 overexpression and postconditioning, is accomplished through the activation of the Akt/GSK3β survival pathway, no synergic effect was evidenced. Thioredoxin-1 plays a key role in the postconditioning, given that when this protein is inactive the cardioprotective mechanism was abolished. Thus, diverse comorbidities or situations modifying the thioredoxin activity, could explain the absence of this strong mechanism of protection in different clinical situations.  相似文献

目的:研究基质金属蛋白酶9(MMP-9)基因多态性与缺血性脑卒中(IS)发病及预后的相关性,为IS的防治提供新的理论依据。方法:选取治疗的IS患者100例,根据TOAST分型标准分为大动脉粥样硬化型(LAA)组41例,小动脉阻塞型(SAO)组59例,并选取健康体检者40例作为对照组,采用PCR-RFLP法检测各组MMP-9基因C1562T、R279Q多态性,并对IS患者进行3个月的随访,采用Logistic回归分析C1562T、R279Q多态性与IS患者预后的相关性。结果:LAA组、SAO组MMP-9基因C1562T位点T等位基因、C/T+T/T基因型频数均高于对照组,差异有统计学意义(P0.05),LAA组、SAO组C1562T位点C等位基因、C/C基因型频数及R279Q位点等位基因和基因型频数与对照组比较差异无统计学意义(P0.05);Logistic回归分析显示,MMP-9各型别基因与预后无明显相关性(P0.05)。结论:MMP-9基因C1562T的T等位基因是IS发病的穿易感基因之一,但MMP-9基因多态性与IS患者的预后并无明显相关性。  相似文献

目的:检测分析急性力竭运动对大鼠心肌蛋白激酶C(PKC)α、δ和ε亚型总蛋白及其磷酸化蛋白表达的影响。方法:健康雄性SD大鼠50只,随机分为对照组(C组)和急性力竭运动组(AEE组)(n=25),采用一次力竭跑台运动建立急性力竭运动模型,Western blot法检测PKCα、δ和ε总蛋白及其磷酸化蛋白的表达水平。结果:与C组比较,AEE组大鼠心肌PKCα、p-PKCαSer-657、PKCδ、p-PKCδThr-507及PKCε水平均显著升高(P<0.05)。结论:急性力竭运动导致PKCα、PKCδ、PKCε表达水平和PKCα、PKCδ活化水平上调,可能是急性力竭运动导致大鼠心肌损伤的重要因素,三亚型之间的交互作用可能是更重要的因素。  相似文献

目的:探讨不同剂量的血必净注射液对缺氧/复氧大鼠心肌功能的保护作用。方法:采用Langendorff方法制备大鼠离体心脏缺氧/复氧模型。130只雄性SD大鼠随机分为对照组(sham组),缺氧/复氧组(H/R组),低、中、高剂量血必净组(XBJL、XBJM、XBJH组),除对照组外,其他四组按复氧不同时相(复氧0.5 h、1 h、2 h)又分别分为3个亚组(n=10)。对照组在平衡灌注20 min时纪录左室发展压(LVDP)、左心室发展压最大上升/下降速率(±dp/dtmax)、左心室内压(LVP)、心率(HR)的值, ELISA检测心肌中肌酸激酶同工酶(CK-MB)的浓度,光镜下观察心肌组织结构的改变;其余各组平衡灌注20 min后,灌注ThomasⅡ停搏液使心脏完全停搏30 min之后复灌K-H液使其心脏复跳,连续记录LVDP、±dp/dtmax、LVP、HR在复氧不同时间点的动态变化,ELISA检测各组复氧不同时间点心肌中CK-MB的浓度,光镜下观察各组复氧不同时间点心肌组织结构的改变。结果:与sham组相比,其余各组LVDP、±dp/dtmax、LVP值均降低(P<0.05),心肌中CK-MB浓度上升(P<0.05),心肌组织结构发生异常改变,随着复氧时间延长,以上指标异常变化逐渐加剧;在复氧0.5 h、1 h、2 h,各剂量血必净组LVDP、±dp/dtmax、LVP的值均高于H/R组对应时间点的值(P<0.05),心肌中CK-MB浓度均低于H/R组,心肌组织结构异常变化减轻,以中剂量改善效果最佳(P<0.05)。结论:血必净注射液能够有效改善缺氧/复氧大鼠心肌的功能及形态学结构,以中剂量血必净(4 ml/100 ml)效果最佳。  相似文献

Right ventricular (RV) dysfunction is a common cause of heart failure in patients with congenital heart defects and often leads to impaired functional capacity and premature death. Myocardial tissue regeneration techniques are being developed for the potential that viable myocardium may be regenerated to replace scar tissues in the heart or used as patch material in heart surgery. 3D computational RV/LV/Patch models with fluid-structure interactions (FSI) were constructed based on data from a healthy dog heart to obtain local fluid dynamics and structural stress/strain information and identify optimal conditions under which tissue regeneration techniques could achieve best outcome. RV/LV/Patch geometry and blood pressure data were obtained from a dog following established procedures. Four FSI models were used to quantify the influence of different patch materials (Dacron scaffold, treated pericardium) on local environment around the patch area, especially focusing on the thickness and stiffness of the patch. Our results indicated that changes in patch stiffness had little impact on the ejection fraction of the right ventricle because the total patch area was small. However, patch stiffness had huge impact on local RV maximum principal stress (Stress-P1) and strain (Strain-P1) around the patch area. Compared to the no-patch model, patch models had increased Stress-P1 and decreased Strain-P1 values in the patch area. Softer patches were associated with greater stress/strain variations. Thinner patch led to complex local flow environment which may have impact on myocytes seeding and RV remodeling. Our multi-physics RV/LV/Patch FSI model can serve as a useful tool to investigate cellular biology and tissue regeneration under localized flow and structural stress environment.  相似文献

The effect of exogenous ATP and its analogs on heart function was studied in 14–100-day-old rats. Extracellular purines had a positive chronotropic effect on the heart. Intravenous administration of exogenous ATP and its stable analogs induced a dose-dependent increase in heart rate depending on animal age. The analysis of isometric contraction of myocardial strips demonstrated a dose-dependent positive inotropic effect of ATP. The family and subtype of the P2 receptors realizing the positive chronotropic and inotropic effects were identified using selective agonists and blockers. P2X receptors demonstrated the highest sensitivity during early postnatal ontogeny. The age-related pattern of the receptor response to exogenous purines indicated the heterochronic maturation of P2X and P2Y receptors in the myocardium.  相似文献

The aim of this study was to determine the relationship between serum and cerebrospinal fluid (CSF) magnesium (Mg+2) levels, Glasgow Coma Scores (GCS), and 7-day mortality in acute stroke patients. Patients with acute ischemic or hemorrhagic stroke arriving within the first 3 h of symptoms were included in the study. The control group consisted of healthy volunteers. GCS was determined, and blood and CSF samples were taken in order to establish serum and CSF glucose, Mg+2, sodium, potassium, calcium, and chlorine levels. Mortality was recorded at 7 days after admission. CSF Mg+2 in the ischemic infarct group was significantly lower than in the control group (p = 0.006). CSF Mg+2 in the ischemic infarct patients with a GCS ≤ 8 were significantly lower (p = 0.002) than controls and in ischemic infarct patients with a GCS ≥9. In the ischemic stroke patients, CSF Mg+2 and GCS were significantly correlated (r = 55, p = 0.031). CSF Mg+2 levels in ischemic stroke patients who died within 7 days were significantly lower than controls, ischemic stroke patients who survived, and hemorrhagic stroke patients who died (p = 0.002, p = 0.042, and p = 0.005, respectively). Low CSF Mg+2 levels in patients with acute ischemic stoke at admission predicted a higher 1-week mortality.  相似文献

On isolated hearts obtained from control rats and rats subjected to regular physical exercises (forced swimming) during 6 weeks, we studied the contractile activity of the heart, resistance of the myocardium to ischemia/reperfusion-induced injuries, as well as the dependence of the developed and end-diastolic pressures in the aortic ventricle (AV) on the strain of the myocardium (by means of a dosed increase in the volume of a polyethylene reservoir inserted into the ventricle). It was demonstrated that adaptation to regular graduated physical exercises exerts a positive effect on the functional state of the AV myocardium and its contractile function. This was manifested in intensification of the contractile activity of the myocardium, a decrease in its oxygen “job cost,” and an increase in the resistance to injuries induced by ischemia-reperfusion. In addition, regular physical trainings led to an increase in the resistance of the AV myocardium to the strain. In trained rats, the plateau of the Frank–Starling plot was significantly greater than that in control animals, while the rigidity of the AV myocardium was significantly lower. 目的:观察大鼠心肌缺血/再灌注损伤对血清和心肌组织瘦素(Leptin)表达的影响,探讨Leptin在心肌缺血/再灌注损伤中的作用。方法:建立大鼠心肌缺血/再灌注模型,检测血清乳酸脱氢酶(LDH)和Leptin浓度,并用HE染色和免疫组织化学观察心肌组织病理学及Lepfin表达水平。结果:缺血组、再灌注组血清LDH水平显著升高(P〈0.05),表明该模型制作成功,造成心肌局部一定程度的损伤。缺血组血清Leptin含量(6.34±2.49)ng/ml显著低于对照组(7.50±2.93ng/ml,P〈0.05);再灌注后Leptin水平缓慢恢复,于再灌注2h时Leptin达到(8.32±1.74)ng/ml,恢复到损伤前水平(8.38±2.56)ng/ml,且随再灌注时间延长有升高趋势。免疫纽化显示与假手术纽心肌Leptin蛋白表达水平相比,其他四组均有显著降低(P〈0.01),按缺血45min后再灌注1h组、缺血45min后再灌注3h组、单纯缺血45min组、缺血45min后再灌注2h组依次递减。结论:Leptin在心肌缺血/再灌注损伤后早期45min血中有明显减少,心肌组织中也明显表达下降。心肌组织病理损伤与Leptin的改变可能有一定的关系。  相似文献

目的:研究雷米普利与BQ-123合用对大鼠在体心肌缺血/再灌注损伤的影响。方法:健康雄性Wistar大鼠随机分为5组,制备缺血30min再灌注120min模型,采用雷米普利、BQ-123单用及两药联合应用的方式处理实验动物。观察两药合用对大鼠在体心肌缺血/再灌注损伤的保护作用。观察动物心率、血压、心电图ST-段变化,记录缺血期室性心律失常;检测血浆CK及LDH活力;心肌HE染色和TIC染色,定性和定量检测心肌梗死情况。结果:与I/R组比较,各给药组ST-段均明显降低;缺血期室性心律失常(VA)出现时间明显推迟且持续时间明显缩短,联合给药组作用更为显著;心律失常发生率明显降低;血浆CK及LDH活力显著降低且联合给药组降低更为显著;梗死面积明显缩小,心肌损伤程度明显减轻,其中联合给药组变化更为显著。结论:雷米普利、BQ-123单用及联合应用均对大鼠在体心肌缺血/再灌注损伤有保护作用,且两药合用在推迟缺血期VA的出现时间,缩短其持续时间,减少CK及LDH漏出,缩小心肌梗死面积方面优于两药各自单用。  相似文献
